چکیده

The newly discovered Ini1 cellular protein binds HIV-1 integrase and is part of a protein complex thought to alter nucleosomal structure; such alterations may influence the selection of sites for HIV-1 DNA integration.

Structural and Functional Role of INI1 and LEDGF in the HIV-1 Preintegration Complex

Integration of the HIV-1 cDNA into the human genome is catalyzed by the viral integrase (IN) protein. Several studies have shown the importance of cellular cofactors that interact with integrase and affect viral integration and infectivity. In this study, we produced a stable complex between HIV-1 integrase, viral U5 DNA, the cellular cofactor LEDGF/p75 and the integrase binding domain of INI1 ...

Structure-function analysis of integrase interactor 1yhSNF5L1 reveals differential properties of two repeat motifs present in the highly conserved region (retroviral integrationyprotein–protein interactionsySWIySNF complexytranscription)

Retroviral integrase (IN) catalyzes the integration of retroviral cDNA into host chromosome. Ini1 (integrase interactor 1) is a host protein that specifically binds and stimulates in vitro joining activity of HIV-1 IN. Ini1 has homology to yeast transcription factor SNF5 and is a component of the analogous mammalian SWIySNF complex that can remodel chromatin. Little is known about the function ...

Functional Coupling between HIV-1 Integrase and the SWI/SNF Chromatin Remodeling Complex for Efficient in vitro Integration into Stable Nucleosomes

Establishment of stable HIV-1 infection requires the efficient integration of the retroviral genome into the host DNA. The molecular mechanism underlying the control of this process by the chromatin structure has not yet been elucidated. We show here that stably associated nucleosomes strongly inhibit in vitro two viral-end integration by decreasing the accessibility of DNA to integrase. Remode...

Structural and functional studies of HIV-1 pre-integration complexes

HIV-1 integrase (IN) is a key component of the pre-integration complex (PIC) and is involved in several steps of retrovirus replication. IN cannot perform these functions on its own and need to recruit host cell proteins to efficiently carry out the different processes. Retroviral INs are flexible proteins showing high inter-domain flexibility. This intrinsic flexibility accounts for IN ability...

Integration of Gender-sensitive Approach to Safe Motherhood Program for the Prevention of STD/ HIV in Iran: A Qualitative Study

Background & aim: Sexually transmitted infections (STIs) present a serious public health burden, which are considered as the factors contributing to acute illnesses, infertility, long-term disability, and mortality. The aim of the present study was to provide an in-depth understanding of the participants' perceptions about the integration of gender-sensitive approach to safe motherhood program ...
